- THIS IS A REQUEST FOR INFORMATION (RFI): The Department of Brain Trauma Neuroprotection and Neurorestoration (BTNN), Center for Military Psychiatry and Neuroscience in support of the Walter Reed Institute of Research (WRAIR), will be conducting studies investigating biomarkers in blood (serum or plasma), cerebral spinal fluid, and brain tissues following mild (concussive) traumatic brain injury (TBI) and penetrating ballistic-like brain injury models of neuronal injury. FACS methodology uses antibody binding to intact, live cells to measure any cell specific biomarker of TBI, sort the cells according to florescent labeling, and collect live sorted cells for later experiments. Description of Services: Engineer from the vendor shall install FACS equipment outlined in the MEC in BTNN, train staff for use, and provide troubleshooting and maintenance. Minimum Essential Characteristics (MEC) of Equipment Possess at least 3 solid state lasers and detect at least 8 fluorophores Have each 488nm /80 mw, 640nm/50 mw, and 405nm/50mw lasers with laser spatial separation and individual focal points which allows for easy alignment and lower compensation (3 pinholes). Configured with 3 colors from a blue laser (488-80), one color from a red laser (638-50), and two colors from a violet laser (405-50) Allow optional forward scatter photomultiplier for small particle detection a small as 100nm Contain a Bubble detector in the sample line detects air bubbles from the sample tube and stops sample flow when the sample tube is empty, preventing air bubbles from reaching the nozzle assembly and comprising sort performance Have real time video monitoring of cell sorting and fluorophore detection Be capable of sorting and collecting live cells Be capable of sorting dead cells into separate fractions and divert to waste Be compatible with standard and deep 96-well plates for analysis and collection Must operate at variable temperature (~10-18 degrees Celsius) to preserve cell viability during cell suspension intake, analysis, and collection Fit onto a standard laboratory bench Be expandable to include an aerosol control chamber Collect cells (have an acquisition rate of) 100,000 events/second Sort live cells with greater than 80% of expected yield and have a two-way sort purity of 98% Be able to fit inside a Biosafety cabinet Generate post-sorting results that can be precisely traced back to the flow characteristics of the specific cell or combinations of cells sorted so you can identify exactly where the cell or cells were sorted from the population of interest. Have the ability to accurately calibrate drop delay Employ live video feed of the droplet breakoff point, waste collection, and side streams Be installed with fully operational workstation and controller PC running Windows 7 operating system and containing instrument and analytical software Capable of sorting single cells for cloning and cell culture development 3-5 days training of operators on site at BTNN, WRAIR 3-5 days operator course training for 1 operator at vendor training site Maintenance service agreements (5 year plan: base year plus 4 option years) for FACS and also for installed lasers to include: oMonday through Friday maintenance service coverage olabor, travel expenses and parts for annual preventive maintenance, and annual preventive maintenance kit oinstallation of software revisions released during the agreement term at no additional cost ounlimited service visits (Monday-Friday excluding holidays) at no additional cost ounlimited telephone support for instruments, reagents and applications at no additional cost Period of Performance: The period of performance will be one year beginning at install date (base year) followed by 4 additional option years for maintenance service contract.
